Glass Beads

Glass beads with a density of 2.5 grams/cc are the primary beads used for “beadbeating.” They come preloaded into self-standing 2 and 7 milliliter tubes as well as one pound bottles, for convenient beadbeating use.

For rapid plating of yeast and bacteria, add a few sterile glass beads to solid nutrient medium in a plate, shake vigorously, then use a Mini-Beadbeater in dry grinding mode at ambient or liquid nitrogen temperatures to quickly pulverize cells into their desired size.

Pre-filled microvials from some vendors allegedly contain unique combinations of sizes and types of beads for improved cell lysis, but without comparative data available the ‘improvements’ may only be marginal. You’d save both money and time by following our guidelines above, loading your vials yourself using commercial or Do-It-Yourself bead loaders; loading only each vial once is enough! When selecting zirconia beads for use in your mill machine, make sure they match up with the size of parts being processed in it.

Silicon carbide (commonly referred to as beta-SiC, -SiC and alpha-SiC) is an extremely hard material which ranks on the Mohs scale between soft alumina (9) and diamond (10). Silicon carbide has become an indispensable material in industrial products that require thermal and mechanical performance, such as abrasives, refractories, ceramics and cutting tools for various industrial uses.

This angular material features sharp points and edges that make it highly effective in rock tumbling, gem cutting and lapidary work of all sorts. Furthermore, its low cost compares favorably with other abrasives like tungsten carbide.

Beadbeating of spores or tough tissue often necessitates using metal beads for more vigorous sample disruption, and Chrome Steel beads with density of 5.5g/cc are ideal. Chemically inert and resistant to fragmentation during beadbeating, these chrome steel beads offer much lower costs and easier access than their stainless steel counterparts.

Cryo-grinding or dry grinding with dense beads requires special reinforced polypropylene microvials or stainless steel screw cap microvials (see XXTuff products), to avoid contamination with rust. BioSpec Products’ chrome steel beads come with an easy way of doing this with their small rare-earth magnet provided in each pack – making the task simpler!

Glass beads differ from stainless steel in that they can be “disposable”, meaning no need for constant cleaning between applications. Only 1 to 3 beads should be placed into each 2 mL screw-cap microvial; typically only 1-3 are needed per application such as dry-grinding leaves and seeds or beadbeating hair; typically these one pound bottles.

Stainless Steel Beads

Stainless steel beads feature a density of 7.9g/cc and are composed of Type 316 stainless, the most corrosion-resistant alloy available. While these expensive beads can be cleaned and reused when used sparingly for dry grinding leaves and seeds, chrome-steel beads have similar features at an enormously cheaper cost, often considered disposable; chrome steel works very well when beadbeating but requires quick retrieval from lysate samples in order to avoid contamination from rust (use a rare-earth magnet for retrieval).

Garnet particles are chemically inert and resistant to fragmentation, unlike SiC or Zirconia which abrade during beadbeating and glass beads, making them superior as tough tissue samples require stronger beads for bead beating. They’re very dense too – often used instead of stainless steel beads when beadbeating for spores or most tissue samples as they accelerate lysis by reducing beadbeating abrasion; like SiC, however, Garnet particles can leave samples with yellow-gray hues; ideal combinations include hard centrrigugation of homogenate homogenate.
